Efficiënte IDUA-genmutatiedetectie met gecombineerd gebruik van dHPLC en gedroogde bloedmonsters
Doelstellingen . Ontwikkeling van een eenvoudige, op mutatie gerichte methode om de kosten van mutatietesten te verlagen met behulp van een gemakkelijk verkrijgbaar biologisch materiaal. Beoordeling van de haalbaarheid van een dergelijke methode werd getest met behulp van een GC-rijk amplicon. Ontwerp en methoden . Een methode voor het denatureren van high-performance vloeistofchromatografie (dHPLC) werd verbeterd en geïmplementeerd als een techniek voor de detectie van varianten in exon 9 van het IDUA-gen . De geoptimaliseerde methode werd getest in 500 genomische DNA-monsters verkregen uit gedroogde bloedvlekken (DBS).
Resultaten. Met deze dHPLC-aanpak was het mogelijk om verschillende varianten te detecteren, waaronder de gemeenschappelijke p.Trp402Ter-mutatie in het IDUA-gen. Het hoge GC-gehalte interfereerde niet met de resolutie en betrouwbaarheid van deze techniek, en discriminatie van GC-transversies werd ook bereikt. Conclusie. Deze op PCR gebaseerde dHPLC-methode is een snelle , gevoelige en uitstekende optie gebleken voor het screenen van talrijke monsters verkregen uit DBS. Bovendien resulteerde het in de consistente detectie van duidelijk te onderscheiden profielen van de gemeenschappelijke p. Trp402Ter IDUA- mutatie met een voordelige balans tussen kosten en technische vereisten.

Vooral in tijden als de huidige COVID-19-pandemie kunnen geavanceerde en schaalbare platformtechnologieën efficiëntere en kosteneffectievere oplossingen bieden om aan de wereldwijde vraag naar vaccins te voldoen. Hierin bespreken we de heersende literatuur over de ontwikkeling van Vero-celbioprocessen voor de productie van virale vectoren en vaccins met als doel de recente vooruitgang in de ontwikkeling van bioprocessen te beoordelen.
We onderstrepen kritisch de noodzaak van verdere onderzoeksactiviteiten en beschrijven knelpunten om het Vero-celplatform te verbeteren door gebruik te maken van recente ontwikkelingen op het gebied van celcultuurtechnologie.
Efficiënte Inductie van cytotoxische T-cellen door Viral Vector Vaccinatie Vereist STING-Dependent DC Functies
Gemodificeerd vacciniavirus Ankara (MVA) is een verzwakte stam van het vacciniavirus en wordt momenteel onderzocht als een veelbelovende vaccinvector tegen infectieziekten en kanker. MVA verwierf mutaties in gastheerbereik en immunomodulerende genen, waardoor het virus deficiënt was voor replicatie in de meeste zoogdiercellen. MVA heeft een hoog veiligheidsprofiel en induceert robuuste immuunresponsen. De rol van aangeboren immuuntriggers voor de inductie van cytotoxische T-celreacties na vaccinatie is echter onvolledig begrepen.
Stimulator van interferon-genen (STING) is een adapter-eiwit dat de signalering stroomafwaarts van verschillende DNA-sensoren integreert en daardoor de inductie van sort I-interferonen en andere cytokinen of chemokinen bemiddelt als reactie op verschillende dsDNA-virussen. Omdat de sort I-interferonrespons volledig STING-afhankelijk was tijdens MVA-infectie, hebben we het impact van STING op primaire en secundaire cytotoxische T-celresponsen en geheugen T-celvorming na MVA-vaccinatie in STING KO-muizen bestudeerd .
Bovendien analyseerden we de impression van STING op de rijping van beenmerg-afgeleide dendritische cellen (BMDC’s) en hun functionaliteit als antigeenpresenterende cellen voor cytotoxische T-cellen tijdens MVA-infectie in vitro . Geschikte methoden voor de productie van een Orf-virus , inclusief up- en downstream, zijn echter zeer beperkt. De gepresenteerde studie richt zich op downstream-verwerking en beschrijft de evaluatie van verschillende chromatografische eenheidsbewerkingen.
In deze context werden ionenuitwisselings-, pseudo-affiniteits- en sterische exclusiechromatografie toegepast voor de zuivering van het uit celkweek afgeleide Orf-virus, met als doel een maximale virusterugwinning en contaminantuitputting. De meest veelbelovende chromatografische methoden voor het vangen van de virusdeeltjes waren de sterische exclusie- of zouttolerante anionenuitwisselingsmembraanchromatografie, waarbij 84% en 86% van het infectieuze virus werd teruggewonnen.
Door de sterische exclusiechromatografie te combineren met een daaropvolgende Capto TM Core 700-hars- of hydrofobe interactiemembraanchromatografie als secundaire chromatografische stap, werd een totale virusterugwinning tot 76% bereikt. Bovendien was een volledige verwijdering van cellulair eiwit en een DNA-depletie van de gastheercel tot 82% mogelijk voor de sterische uitsluitingsmembranen en de Capto TM Core 700-combinatie. De studie onthult een reeks mogelijke eenheidsoperaties die geschikt zijn voor de chromatografische zuivering van het uit celkweek afgeleide Orf-virus, afhankelijk van de beoogde toepassing, dwz een humaan of veterinair gebruik, en de vereiste zuiverheid.

Description: Interleukin 8 (IL-8 or chemokine (C-X-C motif) ligand 8, CXCL8) is a chemokine produced by macrophages and other cell types such as epithelial cells, airway smooth muscle cells and endothelial cells. IL-8, also known as neutrophil chemotactic factor, has two primary functions. It induces chemotaxis in target cells, primarily neutrophils but also other granulocytes, causing them to migrate toward the site of infection. IL-8 also stimulates phagocytosis once they have arrived. IL-8 is also known to be a potent promoter of angiogenesis. In target cells, IL-8 induces a series of physiological responses required for migration and phagocytosis, such as increases in intracellular Ca2+, exocytosis (e.g. histamine release), and the respiratory burst. IL-8 can be secreted by any cells with toll-like receptors that are involved in the innate immune response and has been demonstrated to be a signatory chemokine of CR2+ naive T cells, also known as recent thymic emigrants. Usually, it is the macrophages that see an antigen first, and thus are the first cells to release IL-8 to recruit other cells. Both monomer and homodimer forms of IL-8 have been reported to be potent inducers of the chemokine receptors CXCR1 and CXCR2. The homodimer is more potent, but methylation of Leu25 can block the activity of homodimers. |
