Diversiteit van de microbiota die betrokken zijn bij de productie van wijn en biologische appelcider ondergedompeld azijn, zoals onthuld door DHPLC-analyse en sequencing van de volgende generatie

Ongefilterde azijnmonsters verzameld uit drie oxidatiecycli van de ondergedompelde industriële productie van elk, rode wijn en biologische appelciderazijn, werden bemonsterd in een Sloveens azijnproducerend bedrijf. De monsters werden systematisch verzameld van het start tot het einde van een oxidatiecyclus en gebruikt voor kweekonafhankelijke microbiële analyses die werden uitgevoerd door denaturerende hogedrukvloeistofchromatografie (DHPLC) en Illumina MiSeq-sequencing van 16S rRNA- genvariabele regio’s.

Beide benaderingen vertoonden een zeer homogene bacteriële structuur tijdens de productie van wijnazijn, maar meer heterogeen tijdens de productie van biologische appelciderazijn. In alle wijnazijnmonsters was Komagataeibacter oboediens (voorheen Gluconacetobacter oboediens) een overheersende soort. In appelciderazijn waren de azijnzuur- en melkzuurbacteriën twee belangrijke groepen bacteriën.

Het azijnzuurbacteriële consortium bestond uit Acetobacter en Komagataeibacter, waarbij het geslacht Komagataeibacter de Acetobacter overtrof in alle appelciderazijnmonsters aan het einde van de oxidatiecyclus. Onder het melkzuurbacteriële consortium werden twee dominante geslachten geïdentificeerd, Lactobacillus en Oenococcus, waarbij Oenococcus de overhand had met toenemende concentratie azijnzuur in azijn.

Onverwacht was Gluconobacter een minder belangrijk geslacht van het azijnzuurbacteriële consortium in biologische appelciderazijn , wat wijst op een mogelijke ontwikkeling van de Gluconobacter- populatie met een tolerantie tegen ethanol en azijnzuur. Onder de begeleidende bacteriën van de wijnazijn werd het geslacht Rhodococcus gedetecteerd , maar dit nam aanzienlijk af tegen het einde van de oxidatiecycli

Ontwikkeling van op tomaten-struikachtige stuntvirus   gebaseerde  vectoren  voor fusie en niet-fusie-expressie van heterologe eiwitten in een alternatieve gastheer Nicotiana excelsiana

Op plantenvirus gebaseerde expressiesystemen zijn een alternatief expressieplatform voor de productie van klinisch en industrieel bruikbare recombinante eiwitten . Niettemin, vanwege een gebrek aan virale vectoren met de commerciële mogelijkheden, is het dringend nodig om nieuwe, veelzijdige en efficiënte plantenvirusvectoren te ontwerpen en te ontwikkelen.

Het genoom van Tomato Bushy Stunt Virus (TBSV) biedt een aantrekkelijk alternatief voor modificatie als vector voor het produceren van heterologe eiwitten in planten. Hier hebben we een reeks nieuwe fusie- en niet-fusie- TBSV-CP-vervangingsvectoren ontwikkeld , die flexibelere en efficiëntere hulpmiddelen bieden voor het tot expressie brengen van eiwitten van belang in planten. Een alternatieve tabaksplant, Nicotiana excelsiana, werd in deze studie gebruikt als gastheer voor nieuw geconstrueerde TBSV-vectoren, omdat de ongewenste necrotische effecten werden gerapporteerd op de algemeen gebruikte Nicotiana benthamiana-gastheer geassocieerd met expressie van TBSV-gecodeerd P19-eiwit.

De gegevens toonden aan dat TBSV-vectoren een symptoomloze infectie veroorzaakten en reportergen tot overexpressie brachten in bladeren van N. excelsiana, wat aantoont dat N. excelsiana een ideale waardplant is voor TBSV-gemedieerde heterologe genexpressie. Bovendien toont een TBSV-niet-fusievector, dAUG, het accumulatieniveau van reportereiwitten dat vergelijkbaar is met dat van op TMV en PVX gebaseerde vectoren in zij-aan-zij vergelijking en biedt flexibelere aspecten dan de eerder ontwikkelde TBSV-vectoren. Gezamenlijk voegt ons nieuw ontwikkelde TBSV-expressiesysteem een nieuw lid toe aan de familie van plantaardige virale expressievectoren en biedt het ondertussen een flexibele en zeer effectieve benadering voor het produceren van eiwitten die van belang zijn in planten.

 

BELANGRIJKE PUNTEN:

    • Het op TBSV gebaseerde transiënte expressiesysteem is aanzienlijk verbeterd.

  • De necrotische effecten veroorzaakt door viraal P19-eiwit werden vermeden door het gebruik van N.

excelsiana als waardplant.

  • Het expressieniveau van de niet-fusievector was vergelijkbaar met het meest effectieve virus

tot nu toe gerapporteerde vectoren.

Impact van serotype en Pressure Diversiteit op Dengue  Virus  Replication in Australische Mosquito  vectoren

Dengue-virus (DENV) is de belangrijkste door muggen overgebrachte virale ziekteverwekker van mensen, bestaande uit vier serotypen (DENV-1 tot -4) met een groot aantal genotypen en stammen. De kinetiek van DENV-replicatie in de mug na inname van een bloedmaaltijd beïnvloedt het vermogen van de ziekteverwekker om de speekselklieren te bereiken en daarmee het transmissiepotentieel . De invloed van DENV-serotype en stamdiversiteit op viruskinetiek in de twee belangrijkste vectorsoorten,  Aedes aegypti  en  Ae. albopictus , is slecht gekarakteriseerd. We hebben getest of DENV-replicatiekinetiek systematisch varieert tussen serotypen en stammen, met behulp van Australische stammen van de twee vectoren.

Muggen werden met bloed gevoed met twee stammen per serotype en bemonsterd op 3, 6, 10 en 14 dagen na blootstelling. Virusinfectie in muggenlichamen en verspreiding van virus naar poten en vleugels, werd gedetecteerd met behulp van qRT-PCR. Voor beide vectoren vonden we significante verschillen tussen serotypen in percentages geïnfecteerde muggen, met hogere aantallen voor DENV-1 en -2 versus andere serotypen . In overeenstemming hiermee hebben we waargenomen dat DENV-1 en -2 in het algemeen repliceerden tot hogere RNA-niveaus dan andere serotypen, vooral op eerdere tijdstippen. Er waren geen significante verschillen in infectiesnelheid of verspreiding tussen de muggensoorten. Onze resultaten suggereren dat DENV-diversiteit belangrijke epidemiologische gevolgen kan hebben door de viruskinetiek in mugvectoren te beïnvloeden.

Een Plant  Virus  Zorgt  Viral  Stabiliteit in de hemolymfe van  Vector  insecten door middel onderdrukken profenoloxidase Activation

De meeste plantenvirussen hebben vectorinsecten nodig voor overdracht. Virale stabiliteit in de hemolymfe van vectorinsecten is een voorwaarde voor succesvolle overdracht van persistente plantenvirussen . Of de proteolytische activering van profenoloxidase (PPO) de stabiliteit van persistente plantenvirussen beïnvloedt, blijft echter ongrijpbaar.

Hier hebben we het samenspel tussen rijststreepvirus (RSV) en de PPO-cascade van de vector kleine bruine planthopper onderzocht. De activiteit van fenoloxidase (PO) werd door RSV met ongeveer 60% onderdrukt. Toen de PPO-cascade werd geactiveerd, vonden we duidelijke melanisatie rond RSV-deeltjes en ernstige schade aan de virale stabiliteit in de hemolymfe. Virale onderdrukking van PO-activiteit was afgeleid van obstructie van proteolytische splitsing van PPO’s door binding van het virale niet-structurele eiwit NS3. Deze resultaten geven aan dat RSV de PPO-respons verzwakt om de virale stabiliteit in de hemolymfe van vectorinsecten te verzekeren. Ons onderzoek biedt verhelderende aanwijzingen voor het beheersen van de overdracht van door vectoren overgedragen virussen.

BELANG  Grote hoeveelheden door vectoren overgedragen plantenvirussen circuleren in de hemolymfe van hun vectorinsecten voordat ze de speekselklieren binnendringen om op planten te worden overgedragen.

De stabiliteit van virionen in de hemolymfe is daarbij essentieel. Activering van het proteolytische profenoloxidase (PPO) om actief fenoloxidase (PO) te produceren, is een van de belangrijkste aangeboren immuunroutes in hemolymfe van insecten. Hoe een plantenvirus omgaat met de PPO-immuunreactie in zijn vectorinsect blijft onduidelijk.

Hier melden we dat de PPO de stabiliteit van het rijststreepvirus (RSV), een berucht rijstvirus, in de hemolymfe van een vectorinsect, de kleine bruine planthopper, beïnvloedt. RSV onderdrukt PPO-activering met behulp van viraal niet-structureel eiwit. Zodra het niveau van PO-activiteit is verhoogd, wordt RSV gemelaniseerd en geëlimineerd uit de hemolymfe. Ons werk geeft waardevolle aanwijzingen voor het ontwikkelen van nieuwe strategieën voor het beheersen van de overdracht van door vectoren overgedragen plantenvirussen.
